Begins with association of RNA polymerase & DNA template
- RNA polymerases bind to promoters
- they cannot recognize promoters on their own
- but require help of other proteins (transcription factors)
- TFs particularly important in initiation of eukaryotic gene transcription
- Promoter specifies which strand
